Abstract:
A method of assaying endotoxin accurately to thereby allow a definite diagnosis of gram negative bacterial sepsis, which has been very difficult heretofore, to be conducted rapidly, by adding an aqueous solution of a mixture of a specified surfactant, a compound containing an imidazolyl or amino group, an alkaline earth metal salt and an alkali metal hydroxide to a specimen and then adding a limulus amoebocyte lysate component thereto. An excellent specimen solution can be prepared because an endotoxin adherent to protein, lipid, platelet, etc., can be liberated efficiently merely by adding the aqueous solution which need not be subjected to various operations including separation. Therefore this method serves to solve the problem of the prior art method of assaying endotoxin by the limulus test using plasma, serum, etc., as a specimen that it necessitates pretreatment such as centrifugal separation of denatured precipitates formed by an acid treatment, so that the operation is complicated.
