Abstract:
To provide a nucleic acid detection method with which nucleic acids can be detected conveniently at high sensitivity and an assay kit.SOLUTION: An embodiment of a nucleic acid detection method is for detecting a target nucleic acid in a sample in which the target nucleic acid includes a first sequence. The method comprises: (A) putting, in the presence of an electrode, a reaction liquid that comprises the sample, a primer set, amplification enzyme, magnesium ions at a concentration of 4 mM to 30 mM, and a ruthenium complex redox probe that generates a detectable electrical signal that increases in strength as the amount of amplification product increases in the reaction field and forming a reaction field in which the nucleic acid probe is not immobilized; (B) keeping the reaction field under amplification reaction conditions; (C) detecting electrical signals from the redox probe over time while keeping the reaction field under amplification reaction conditions; and (D) determining the existence or amount of the target nucleic acid on the basis of the strength of the electrical signal over time obtained in (C).SELECTED DRAWING: Figure 1
