Abstract:
PURPOSE: To obtain a new 7-octyn-1-ene derivative useful as a synthetic intermediate for la-hydroxyvitamin D derivative having a substituent at 2-site by a relatively short reaction process using an easily available inexpensive raw material as a starting material.
CONSTITUTION: The 7-octyn-1-ene of formula I [R1 to R3 are H or OH-protecting group; R4 is H, trisubstituted silyl, CO2R5 or CH2OR6 (R5 is H or alkyl; R6 is H or OH-protecting group), e.g. (5R, 6R, 7S)-tetrahydro-2-(5-hydroxy-6,7-O- isopropylidene-8-nonen-2-yn-1-yloxy)-2H-pyran. The compound of formula I can be produced by reacting an oxysilane derivative of formula II with an acetylene derivative of formula III in the presence of a basic substance or a Grignard reagent and optionally protecting or deprotecting the hydroxyl group of the product.
