Abstract:
PURPOSE: To prepare an agar medium capable of obtaining a measured value having good reproducibility, containing a trace amount of galactose and useful for measuring galactose amount and galactose-1-phosphoric acid amount on the basis of a size of growth band of Escherichia coli by using Escherichia coli and phage.
CONSTITUTION: This ager culture medium is obtained by dissolving a powdery culture medium for measurement of galactose composed of ferric chloride, gelatin, ammonium chloride, magnesium sulfate, glycine, sodium chloride, sodium glutamate, L-methionine, L-threonine, tris(hydroxymethyl)aminomethane hydrochloride, calcium chloride, thiamine, potassium phosphate and lactose into water and adding glycerol, sodium hydroxide, triphenyltetrazolium chloride, an agar agar from which free galactose is removed and Escherichia coli and phage thereto, and further mixing these components with a trace amount of galactose to provide the objective agar culture medium. The agar culture medium is used for a method for measuring galactose amount and/or galactose-1-phosphoric acid amount in blood based on the size of growth zone of Escherichia coli.
