Abstract:
PROBLEM TO BE SOLVED: To provide anti-HER3/HER4 antigen binding proteins, e.g. anti-HER3/HER4 antibodies, that bind to the beta-hairpin of HER3 and the beta-hairpin of HER4 both being hidden epitopes of the receptors, methods for selecting these antigen binding proteins, and methods of their preparation and use as medicament.SOLUTION: The invention provides an antigen binding protein that binds within a specific amino acid sequence of PQPLVYNKLTFQLEPNPHT of human HER3 and a specific amino acid sequence of PQTFVYNPTTFQLEHNFNA of human HER4. The antigen binding protein binds to β hairpin region of HER3 and/or binds to HER3-ECD with a ratio of the association constant in the presence of heregulin (Ka+) and the association constant in the absence of heregulin (Ka-)(Ka+/Ka-) being 4.0 or higher, and binds to β hairpin region of HER4 and/or binds to HER4-ECD with a ratio of the association constant in the presence of heregulin (Ka+) and the association constant in the absence of heregulin (Ka-)(Ka+/Ka-) being 20.0 or higher.SELECTED DRAWING: Figure 2
