Abstract:

PURPOSE: To quickly, and easily characterize virus through such procedures that the RNA of a virus is transformed to the cDNA which, in turn, is amplified in the presence of a primer in the identical sequence region and a restriction pattern examined using various restriction enzymes is compared with those of known viruses.

CONSTITUTION: A virus is characterized through the following procedures: the RNA of the virus is extracted by a conventional means; using the RNA as template, it is transformed to the cDNA by the use of a reverse transcriptase, and the cDNA is amplified by PCR in the presence of primers 1 and 2 in the identical sequence region derived from e.g. the 1st and 2nd collective sequences of rhinovirus; the location of the resulting amplified cDNA on the gene map is determined using restriction enzymes such as Ban II, Hind III, Rsa I, EcoR I, Bgl II, Pvu II, Dra III, and HinP I, and the resultant restriction pattern is compared with those of known viruses.
