Abstract:
To provide a composition and a method for inducing the targeted disruption of endogenous genes within eukaryotic cells.SOLUTION: The present invention provides a composition for inducing the targeted disruption of endogenous genes within eukaryotic cells, including a Cas9/RNA complex. The Cas9/RNA complex includes a Cas9 protein bound to one or more tag sequences, wherein the tag sequence includes a nuclear localization signal (NLS)) and a guide RNA having CRISPR RNA (crRNA) and trans-activating crRNA (tracrRNA), wherein crRNA includes (i) a part that hybridizes with a part of tracrRNA and ii) a part complementary to the target DNA of the endogenous gene. Prior to its introduction into eukaryotic cells, the Cas9 protein is complexed with the guide RNA to form a ribonucleoprotein. The ribonucleoprotein serves as an endonuclease that induces the targeted disruption of the target DNA.SELECTED DRAWING: Figure 1a
