Abstract:

PROBLEM TO BE SOLVED: To provide a method for producing an immunological reagent encoding a cell surface antigen of human origin.
SOLUTION: This method for producing an immunological reagent comprises the following steps: an established non-human oncogenic cell system is transduced with a DNA isolated from human oncocyte and a DNA encoding a selectable or identifiable character, thus transduced cells are selected and collected, and then injected into appropriate 1st rat hosts, the resultant hosts are maintained for a period effective for forming a tumor in the rat hosts by inducing the injected transduced cells, oncocytes are obtained from the resultant tumor and subjected to solid phase formation of an antiserum produced with respect to the above-established non-human nononcogenic cell system, the resultant antiserum solid phase-formed cells are then injected into appropriate 2nd hosts, the resultant hosts are then screened to identify a host capable of forming serum reactive to human oncocyte, the spleen is extracted from the thus identified 2nd host to prepare hybridoma and the hybridoma cell system capable of specifically recognizing a cell surface antigen and producing an antibody bindable thereto is collected.
