Abstract:
PROBLEM TO BE SOLVED: To fill a chromatography column by providing a container containing a medium to be filled, and connected to a side face opening through a pipe line passage and/or to a plunger through a pipe line passage.
SOLUTION: After opening a column outlet port, a filling pump 10 feeds a medium 9 to be filled, into a chromatography column 1 through a side column opening 5, a piston flit 2, and a bottom flit 4 and a pipe line passage (c) of a circulation passage. Next, with the use of suspended RP silica gel 7 is gradually added into the solvent circulation passage by means of a pump 11. Further, a mixture of silica gel and n-propanol is fed into the column 1 through a filling pipe line passage (a). Thus, the chromatography filling can be made by a filtering pressure, and accordingly, the pressure during the chromatography filling can be lowered. As a result, the surface can be made to be flat during filtering, thereby it is possible to make the density distribution of the chromatography filling.
