Abstract:

To obtain a new protein having a structure of a protein having a specific amino acid sequence or a structure wherein the amino acid sequence is partially modified, and useful for the clarification of the functions of human 26S proteasome, the diagnosis and therapy of various diseases such as Alzheimer's disease, etc.

This human 26S proteasome-constituting component P100 protein is a protein having an amino acid sequence of the formula or having an amino acid sequence produced by substituting, removing or adding one or more amino acids of the amino acid sequence of the formula. The protein is useful for the clarification of the functions of human 26S proteasome, for the cause clarification, diagnosis, therapy, etc., of Alzheimer's disease, etc. The protein is obtained by separating mRNA from a human kidney, etc., producing the human 26S proteasome by a conventional method, producing a cDNA library with the mRNA, cloning the produced cDNA library subjecting to PCR with a synthetic DNA coding the N-terminal and C-terminal amino acid regions of human P100 protein, and subsequently expressing the obtained clone in a host cell.
