Abstract:
NEW MATERIAL:A human hepatic parenchymal cell growth factor (hHGF), having an amino acid sequence on the N-terminal side expressed by the formula (Xaa is pyroglutamic acid residue) and a specific amino acid sequence. USE:A therapeutic agent for hepatopathy. PREPARATION:For example, a cDNA of a human hepatic parenchymal cell growth factor (hHGF) is prepared according to a well-known method, treated with a restriction enzyme, linked to a plasmid vector and then transduced into Escherichia coli. Transformation is carried out and the resultant transformant is cultured and proliferated to recover and purify the resultant plasmid DNA. The obtained plasmid DNA is subsequently treated with a restriction enzyme, bound to an expression vector and transduced into an animal cell such as CHO cell to perform transformation. The resultant transformed animal cell is then cultured to express the plasmid DNA. Thereby, the objective hHGF having the amino acid sequence at the N-terminal expressed by the formula and the specific amino acid sequence is obtained.
