Abstract:

PURPOSE: Enzymes or cell bodies are included in regenerated fibroin and formed into a microporous structure to enable easy and simple preparation of the titled immobilized enzyme with high enzymatic activity, high stability and safety.

CONSTITUTION: The objective immobilized enzyme is obtained by including enzymes such as arginase or cell bodies in a microporous structure of regenerated fibroin of at least 50,000mol.wt. which contains at least 50wt%, preferably at least 80wt% of hot water-insoluble fibroin, has a porosity of more than 0.4cc/g of less than 10μ pores and can pass through enzymes or cell bodies out to the system. Or the enzymes or cell bodies are mixed with a silk fibroin solution, stirred at a high speed to load a shearing stress more than 50/sec to allow the silk fibroin occluding the enzymes or cell bodies to coagulate in a porous form, subsequently a salting-out reagent such as sodium chloride is added to crush the porous fibroin, then, the crushed product is rinsed with water and freeze-dried to give the objective immobilized enzyme.
