Abstract:

PURPOSE: To obtain an immobilized enzyme, readily recoverable by magnetic force and capable of promoting the movement of a substrate and its contact with an enzyme by vibration and rotary motion with en external magnetic field and increased in apparent activity by binding the enzyme to the surface of a magnetically anisotropic carrier particle.

CONSTITUTION: The objective immobilized enzyme is obtained by adding magnetically anisotropic carrier particles (e.g. Sr ferrite hard magnetic powder) into an aqueous solution of albumin, making the resultant mixture solution flow into water just after boiling while stirring the water, allowing the mixture to cool as it is for about 1hr, collecting the hard magnetic powder with a magnet, decanting the powder, providing the magnetically anisotropic carrier particles having many amino groups present thereon, adding a 50% aqueous solution of glutaraldehyde to the magnetically anisotropic carrier, separating and washing the carrier, subsequently adding an aqueous solution containing an enzyme (e.g. a glucoamylase) thereto, allowing the mixture to stand at 30°C for 1hr while applying an alternating magnetic field thereto and then washing the resultant immobilized enzyme with water. This immobilized enzyme is characterized by a local presence of the enzyme on the surface of the carrier and a high utilization efficiency of the enzyme and a high rate of its enzymic reaction.
