Abstract:

PURPOSE: To obtain new indole and dihydroquinoline derivs. having potassium channel activating action and useful as antiarrhythmic drugs and anti-ischemic drugs, in particular anti-ischemic drugs and usable also as cardiovascular agents, etc.

CONSTITUTION: The indole and dihydroquinoline derivs. are compds. of formula I [where X is O, a group of formula II or a single bond; R1 is CN, COOR, etc.; R is H, an alkyl, etc.; R3, R3', R4 and R4' are H, an alkyl, etc., but when R3 and/or R4 is not H, R3'=R4'=H; R5 is CN, NO2, CF3, etc.; R6 is H, an alkyl, O-alkyl, NH2, etc.; R7 is H, COOR, etc.; R8 is H, a halogen, etc.; (m) is 0 or 1; and (n) is 1, 2 or 3], e.g. trans-4-(3-cyano-1H-indol-1-yl)-3,4-dihydro-3- hydroxy-2,2-dimethyl-2H-1-benzopyran-6-carbonitrile. The compds. of the formula I (where R2 is OH) are prepd. by allowing compds. of formula III to react with compds. of formula IV in a solvent sch as DMF in the presence of a base such as potassium carbonate.
