Abstract:
PROBLEM TO BE SOLVED: To obtain a new lipid A analogue of low toxicity having beneficial physiological activity, and useful as e.g. a medicine or lead compound therefor, by chemical synthesis from chitobiose or N-acetyl chitobiose as starting material.
SOLUTION: This new lipid A analogue with chitobiose as the basic skeleton is expressed by the formula [R is of the formula CH3(CH2)nCO ((n) is an integer of 8-14)], being low in toxicity, having excellent physiological activity similar to that of lipid A, and being useful as a medicine or lead compound therefor. The lipid A analogue is obtained by the following process: using chitobiose or N-acetyl chitobiose as starting material, its hydroxyl group is acetylated, and through an oxazoline derivative, a benzyl glucoside form is obtained, and the acetyl groups other than the N-acetyl group on the 2-site of the benzyl glucoside form are eliminated, followed by protecting the resultant hydroxyl groups with benzylidene groups and tritylating the hydroxyl group on the 6-site, and deacetylating the N-acetyl group and then acylating the amino group and hydroxyl group on the 3-site followed by deprotection of these groups.
