Abstract:

PURPOSE: To enable the control of the pH and the dissolved oxygen concentration (DO2) preventing the mutual interaction of the values, by changing the flow rates of N2 and O2 together with the flow rate of CO2 to be controlled its pH value, and changing the flow rate of CO2 together with the flow rates of O2 and N2 to be controlled its DO2.

CONSTITUTION: Cell is cultured in a culture vessel 1 under the control of the feed of N2, O2 and CO2 to the gaseous phase part 3 of the vessel 1, keeping the pH value and the DO2 in the culture liquid to respective specific levels. When the pH value is deviated from the preset value, the pH is controlled by adjusting the flow rate of CO2, and at the same time, the change in the O2 partial pressure in the gaseous phase part 3 caused thereby is compensated by controlling the flow rates of N2 and O2. On the other hand, when the DO2 in the culture liquid 2 is deviated from the preset level, the DO2 is controlled by adjusting the flow rates of N2 and O2 without performing the control of the flow rate of CO2.
