Abstract:
To improve the sensitivity of genetic analysis or improve detection efficiency for genetic mutations, by selectively purifying primers having modified molecules at their ends.SOLUTION: Provided is a method for purifying a target nucleic acid having a modified molecule at its end, in a method for detecting a target nucleic acid or determining a target nucleic acid based on the mobility of the nucleic acid in electrophoresis, the method comprising: subjecting a double-stranded nucleic acid having a modified molecule at the end of one of the nucleic acid strands to a heat denaturation treatment to form a single-stranded nucleic acid, and treating the resulting single-stranded nucleic acid with an exonuclease at a heat denaturation temperature to remove nucleic acids other than the nucleic acid strand having the modified molecule at the end.SELECTED DRAWING: Figure 5
