Abstract:
NEW MATERIAL:A compound of formula I (RaWRc, Ra'WRc' are lower alkyl).
EXAMPLE: (4R)-1-(t-Butyldimethylsilyloxyethyl)azetidine-2-one.
USE: β-Lactam antibiotic.
PREPARATION: A compound of formula IV resulting from trialkylsilylation of a compound of formula III (R2 is lower alkyl) such as (4S)-methoxycarbonylmethyl- azetidin-2-one is reduced into a compound of formula V, which is further subjected to trialkylsilylation to give a compound of formula VI. A trialkylsilyl halide is made to act on the product in the presence of lithium diisopropylamide and the product is made to react with a compound of CH3-CO-CH2-OR1 (R1 is hydroxyl-protecting group) to give a compound of formula VII, finally protecting groups are removed.
