Abstract:

PURPOSE: A novel DNA that is obtained by converting a single-stranded DNA in bean golden mosaic virus into a double-stranded DNA, thus being used in gene recombination of plants and a hybrid DNA which is obtained by integrating the novel DNA to vector for other organisms.

CONSTITUTION: The objective novel DNA is prepared by making a single- stranded DNA in bean golden mosaic virus, a kind of gemini viruses, into double-stranded DNA which is easy to handle in genetic engineering. For example, a single-stranded DNA with 258 base units from the above-stated virus is made double-stranded at least partially to give the objective DNA. The resultant double-stranded DNA is broken with restriction enzymes and the vector for and organism which has been broken with the same enzyme is recombined therewith to give the objective hybride DNA. The hybrid DNA is allowed to return back into the original host such as Escherichia coli or yeast and proliferation is carried out to give the hybrid DNA.
