PURPOSE: To obtain new oxyalkynes which inhibit 5-lipoxygenase or 5- and 12-lipoxygenase without affecting cyclogenase activity.
CONSTITUTION: Compounds of formula I [Y is a group of formula II at meta- or para-position (R1 is CH3 or NH2; R2 is H or CH3); R3 is H, a 1-3C alkyl or COCH3; Ar is a group of formulae III-VI (R4-R6 are each H, F, Cl, Br, 1-6C alkyl, CF3 or 1-6C alkoxy; R7 is H, F, Cl, Br, 1-6C alkyl or CF3)], such as N-hydroxy-N-[4-[1-hydroxy-3-phenyl-prop-2-yn-1-yl]-benzyl]acetamide, are provided. The compd. of the formula I is prepared by reacting an alkyne of the formula Ar-C≡CG with a compd. of formula VII to obtain an acetal of formula VIII, converting the acetal into a hydroxyaldehyde of formula IX in a solvent, converting the hydroxyaldehyde into an oxime of formula X, reducing the oxime with a boron-contg. reductant in the presence of an acid and optionally 1-3C alkylalcohol to obtain a hydroxyamine, and converting the hydroxyamine into the titled compd.
