Abstract:

PROBLEM TO BE SOLVED: To obtain a pertussis vaccine that causes no coagulation of antigen protein on the inactivation, and is safe to human body and stable by admixing catechins as an inactivating agent for pertussis physiologically active substances.
SOLUTION: This vaccine includes a catechin as an inactivating agent for pertussis physiologically active substances. In a preferred embodiment, the catechin is obtained by extracting it from raw tea leaves, green tea, oolong tea or black tea with water, hot water, a variety of alcohols or ethyl acetate, and separating and purifying the extract. In the vaccine, the concentration of the catechin is adjusted to 0.01-10 mM. The objective pertussis vaccine is prepared by mixing the pertussis physiologically active substances with the catechin to effect the inactivation reaction, dialyzing the reaction mixture to remove the catechin and obtain pertussis toxoid and allowing the pertussis toxoid to adsorb to the adjuvant. An new vaccine having excellent pertussis immune effect can be provided thereby.
