Abstract:

PURPOSE: To obtain a photochemical labeling reagent useful for genetic phatogonomy by adding a digoxigenin derivative, a spacer molecule, and a furocoumarin derivative to the reagent.
CONSTITUTION: A labeling reagent expressed by a formula, Dig-S-Fu, is synthesized. In the formula, Dig, S, and Fu respectively represent a digoxigenin derivative used as a hapten, a spacer molecule, and a furocoumarin derivative used as a photochemically bindable structure The digoxigenin derivative Dig is a chemically modified stroid derivative and contains a C-3-hydroxyalkyl amine group which is modified with a chemically binclable substituenl The spacer S is composed of polyalky amine, polyethylene glycol, or their combination. The photochemically bindable structure is especially furocoumarin, angelicin (isopsoralen) or psoralen, and their derivatives which photochemically react to nucleic acid. The binding order of the digoxigenin, spacer, and furocumarin can be decided arbitrarily. Usually, the Dig and S are first bound with each other and the Dig-S is made to react to the Fu. It is also possible to first form Fu-S and the Fu-S is made to react to the Dig.
