NEW MATERIAL:A plasminogen-activating agent obtained from the bile of pigs, and having the following characteristics: molecular weight, 80,000±10,000; isoelectric point, 7.1; UV absorption spectrum, λmax=278nm; fibrin-decomposition activity, positive; Ca-containing standard fibrin plate dissolution activity, positive; heat stability, 50% deactivation in 10min. at 80°C, and 30W40% deactivation in 30min. at 60°C; solubility, soluble in physiological saline water; decomposition of synthetic substrate, slightly decomposes Glutaryl-Gly-Arg-MCA, fairly decomposes Pro-Phe- Arg-MCA.
USE: Remedy for thrombosis. Free of side effects. High thermal stability.
PROCESS: The agent is prepared by (1) adding a water-miscible organic solvent to the bile of pigs, (2) extracting the resulting precipitate with a buffer solution at pH7.0W8.0, (3) fractionating the extract with acetone, (4) dissolving the precipitate in a buffer solution, and (5) treating the solution with an ion exchange resin or a hydrophobic absorption chromatography.
