To provide a new method for quickly and accurately determining the nucleotide sequence of a nucleic acid by a positional sequence determination method by hybrid formation with overcoming problems and defects relating to conventional methods and designs.
The new method is to reproduce an array of a single chain probe on a solid support and comprises processes for (a) synthesizing a nucleic acid array including a constant sequence having a length C on the 3' terminal and a random sequence having a length R on the 5' terminal, (b) immobilizing the array to a first support, (c) synthesizing a set of nucleic acids including a sequence compensating with the constant sequence, (d) hybridizing the nucleic acid in the set with the array, (e) extending the nucleic acid of the set with an enzyme by using the random sequence of the array as a template, (f) modifying the extended the set of the nucleic acids and (g) fixing the modified nucleic acid of the set on a second solid support to form the reproduced array of the single chain probe.
