Abstract:

PURPOSE: To easily obtain a specimen for genetic analysis in a short time using a paraffin-embedded preparation by removing paraffin from the paraffin- embedded preparation and heat-treating an aqueous suspension containing the paraffin-free preparation and a surfactant having protein-denaturating action at a specific temperature.
CONSTITUTION: An aqueous suspension containing a preparation produced by removing paraffin from a paraffin-embedded preparation and a surfactant having protein-denaturating action (e.g. sodium dodecylsulfate) is heat-treated at ≥60°C and treated with a proteinase (e.g. papain). The reaction liquid is reacted with an alcoholic aqueous solution containing an organic compound having protein- denaturating action (e.g. hydroxybenzoic acid) to effect the precipitation of nucleic acid chain. This process enables the easy treatment of a paraffin- embedded preparation for the preparation of a specimen for genetic analysis and capable of easily and quickly getting high-purity nucleic acid chain suitable for the gene amplification reaction by polymerase chain reaction (PCR) process, etc.
