PURPOSE: To obtain the titled antibody useful for the diagnosis of tumor, etc., economically on an industrial scale, by transforming E.coli with an N-myc gene fragment cloned from a fibroblastoma cell strain, preparing a fused protein having high stability and antigenicity using said transformed E.coli and using the fused protein as an antigen.
CONSTITUTION: A gene fragment of N-myc prepared by the cloning of a human fibroblastoma cell strain is inserted thrice between CII and N of a vector capable of producing a fused protein of CII and N and produced by utilizing a phase λPL promoter. An E.coli transformed with the product is cultured to obtain a fused protein having high stability and antigenicity. A rabbit is immunized by administering the protein as an antigen and the serum is collected from the rabbit to obtain an anti-N-myc antiserum. The objective antibody can be produced by purifying the antiserum and separating an IgG fraction therefrom.
