Abstract:
PURPOSE: To obtain an optically active alcohol having high optical purity by using a metal borohydride easily available on an industrial scale at a low cost as a hydrogen source.
CONSTITUTION: The objective optically active alcohol is produced by reacting (I) an asymmetric reducing agent derived from (A) an optically active amino alcohol of formula [R1 is H, a lower alkyl or a (substituted)aralkyl; R2 to R5 are each H, a lower alkyl, a (substituted)aryl or a (substituted)aralkyl] (e.g. norephedrine), (B) a metal borohydride (e.g. sodium borohydride) and (C) a Lewis acid (e.g. zinc chloride) or (D) a lower dialkyl sulfate (e.g. dimethyl sulfate) with (II) an asymmetric ketone of preferably formula, R6-CO-R7 [R6 and R7 are each a (substituted)alkyl, a (substituted)aryl or a (substituted)aralkyl], e.g. 2-bromo-3'chloroacetophenone. The amounts of the component A and the component B are 0.01-0.5mol and 0.3-3mol (in terms of borane) based on 1mol of the component E, respectively.
