PURPOSE: To purify and isolate a new compound, zopfiellin, having an antimicrobial activity, etc., in high purity by dissolving the culture solution of a Zopfiella curvata No.37-3 strain in an alkali aqueous solution, washing the solution with an organic solvent, adding an acid to the washed solution, and extracting the mixture with an organic solvent.
CONSTITUTION: This method for purifying a new compound, zopfiellin, of the formula comprises culturing a Zopfiella curvata No.37-3 strain in a seed medium at 25°C for 5 days, inoculating the obtained culture solution on a fermentation medium, rotation- culturing the inoculated strain at 25°C for 12 days, filtering off the cells from the culture solution, adjusting the culture filtrate to a pH of 3 with a 2N hydrochloric acid, extracting the culture filtrate with ethyl acetate to obtain a crude culture product, vacuum-concentrating the extract, adding methanol and a 50% potassium hydroxide aqueous solution to the obtained crude culture product, stirring the mixture at room temperature for 2hr, vacuum-concentrating the mixture, adding water, successively washing the mixture with organic solvents such as hexane and ethyl acetate, cooling the alkali aqueous solution with ice water, adding an acid such as 10% hydrochloric acid, and subsequently extracting the obtained acidic aqueous solution with an organic solvent such as ethyl acetate.
