PURPOSE: To obtain a reagent for immunological analysis by immobilizing an antibody or antigen corresponding to the antigen or antibody in a reagent onto a liposome which is subjected to a dissolution effect by complement activity and by sealing a labeling material consisting of a water soluble metallic indicator or water soluble dye into the liposome.
CONSTITUTION: A desired lipid and crosslinking agent are brought into reaction to introduce the lipid molecules of the functional group which can be conjugated with the antigen or antibody to be immobilized onto the liposome. The resulted functional lipid and cholesterol are put into a flask and a solvent is added thereto to dissolve the same and thereafter the solvent is distilled and sucked and the remaining solid is dried. The aq. soln. of the prescribed labeling material is added into the flask on the wall surface of which the thin film is formed. The flask is stoppered and shaken by which the suspension of the lipsome is obtd. On the other hand, the antigen or antibody to be immobilized to the liposome and the crosslinking agent are brought into reaction in a buffer soln. to introduce the crosslinking group therein, by which the modified antigen or antibody is obtd. The liposome and the modified antigen or antibody are brought into reaction in a suitable buffer soln., by which the reagent for immunological analysis is obtd.
