Abstract:

NEW MATERIAL:A recombinant DNA having an amino acid sequence of formula I (A is Ser or Ala; X is Asp or Glu; Y is Ala or Ser; B is Leu or Met), produced by integrating a glucose dehydrogenanse-coding DNA originated from Bacillus megaterium into a vector for the introduction of DNA into E. coli and replicative in E. coli.

USE: Production of glucose dehydrogenase.

PREPARATION: The amino acid sequence of the amino acid terminal of a purified enzyme prepared by the culture of Bacillus megaterium is determined. An oligonucleotide having a DNA base sequence of a gene estimated from the amino acid sequence is synthesized and is labeled with 32P to obtain a DNA probe. A colony of E. coli having complementarity to the DNA probe is selected and separated. The plasmid DNA is separated from the cell and integrated into a vector for the expression of gene of an E. coli based on the DNA base sequence.
