Abstract:

To provide a method of making a reconstructed artificial tissue having the versatility to permit the simple, rapid and efficient making of the desired artificial tissue, and the reconstructed artificial tissue, such as a tissue model and an organ model, which has a cell layer having the barrier function intrinsic to organisms and a basement membrane structure and can be advantageously applied to pharmacological tests, toxicity tests, etc., of chemical substances, etc.

The reconstructed artificial tissue is obtained by seeding and culturing the desired homogeneous or heterogeneous cells having basement membrane formability on the basement membrane specimen made by treating the cells which are bonded onto a support across the basement membrane and have the basement membrane formability with 0.1% triton X-100 to dissolve and elute the lipid components of the cells and simultaneously dissolving the protein existing on the surface of the basement membrane by using a liquid mixture composed of coexisting 50 mM NH3 and proteolytic enzyme inhibitor and suppressing the decomposition of the basement membrane by the activity of endogenous protease.
