Abstract:

PROBLEM TO BE SOLVED: To obtain a scanning optical measuring apparatus in which the adjustment of a data input signal can be automated by a method wherein a data-acquisition start position in which the difference between data on scanning operations in different scanning directions becomes minimum is detected and the data-acquisition start position of the data input signal is corrected.
SOLUTION: Light from a laser light source 1 is reflected by a galvanomirror 3 and an optical-path changeover mirror 5 so as to be image-formed on a sample 7. As a result, fluorescence which is radiated by the fluorescent pigment of a cell in the sample 7 is advanced backward in an incident optical path, and it is transmitted through a dichroic mirror 3 so as to be condensed by a photomultiplier tube(PMT) 15. A signal processing circuit 20 fetches image data from the output of the PMT 15 on the basis of an input signal in which a data-acquisition start position is set, and a scanning image is created by a computer 22. At this time, a main-body control part 24 controls a scanning stage 17 and the galvanometer 3 according to a scanning waveform, and the sample 7 is scanned two-dimensionally by a laser spot. Then, the computer 22 sets a data-acquisition start position in which the difference between data from two scanning lines becomes minimum, and the data-acquisition start position of the input signal is corrected.
