The specimen inspection preparation method includes: an alignment step in which a certain site of a sealed specimen container containing a specimen, a pestle and a reagent stably supported at certain positions of a container support portion and a needle attached to a syringe are automatically aligned on an axial line of the needle; a penetration step in which the needle is penetrated through a certain site of the specimen container by automatically relatively moving the certain site of the specimen container and the needle after the alignment on the axial line of the needle; and a reagent removal step in which the reagent is automatically aspirated and removed by the penetrated needle or a reagent-dedicated needle newly inserted into a through-hole formed by penetrating the needle.
