Abstract:

To provide a method and composition for amplifying nucleic acid sequences, more specifically, from nucleic acid sequences of pathogens by targeted whole genome amplification.

A method of targeted whole genome amplification for identifying pathogens, comprising the step of amplifying at least one pathogen genome using selected multiple targeted whole genome amplification primers, thereby elevating the quantity of nucleic acid representing the at least one pathogen genome, in relation to the quantity of nucleic acid representing the at least one background genome, the step of producing one or multiple amplification product representing bioagent identifying amplicon from the amplified pathogen genome, the step of measuring the molecular weight of the amplified product by mass spectrometry, and the step of calculating base composition of the amplified product from the molecular weight, thereby identifying the pathogen in sample.
