Abstract:

To overcome the problems associated with the use of biologics, specifically decoy receptors, and to satisfy the existing needs within the art, namely to provide reduced side-effects of biopharmaceutical polypeptides, such as decoy receptors and biologics (biopharmaceuticals) in general, as well as, optimized therapeutic efficacies and significantly improved pharmacokinetics.SOLUTION: Provided are: a population of purified extracellular vesicles (EVs), being substantially free of vesicle aggregates; an EV purified using a combination of a filtration step and a liquid chromatography (LC) step; and a pharmaceutical composition or a kit of parts comprising the population of EVs or the EV. A method for purifying EVs comprises: the steps of: (i) subjecting a medium containing EV to filtration; and (ii) subjecting a sample obtained from step (i) to LC. An in vitro method for increasing the production yield of extracellular vesicles comprises a step of exposing a vesicle-producing cells from which the extracellular vesicles are to be obtained to at least one autophagy inhibitor. An in vitro method for enriching metabolically active and/or anti-apoptotic proteins in extracellular vesicles, comprises a step of exposing the vesicle-producing cells from which the extracellular vesicles are to be obtained to stress-inducing conditions.SELECTED DRAWING: None
