Abstract:
To provide materials and methods for conjugating a water soluble polymer to a therapeutic protein.SOLUTION: A method of preparing a therapeutic protein conjugate comprises the step of contacting a therapeutic protein or a biologically-active fragment thereof with a thiol reductant and a water soluble polymer, or functional derivatives thereof, under conditions that (a) a reduced cysteine sulfhydryl group is produced on the therapeutic protein and (b) the conjugation of the water-soluble polymer to the reduced cysteine sulfhydryl group is allowed. The therapeutic protein has an amino acid sequence with no more than one accessible cysteine sulhydryl group.SELECTED DRAWING: None
