Abstract:
Disclosed are an application of a marker gene in detecting multipotent stem cell residues, a detection method and a kit. The marker gene comprises at least one of TDGF1 and TRIML2, and is a gene with high-specificity expression in human pluripotent stem cells. The screening of the marker gene is based on a high-throughput RNA sequencing technology. Genes with significantly up-regulated specific expression in pluripotent stem cells are screened from the pluripotent stem cells and derivative cell samples of the pluripotent stem cells, a group of candidate genes which can be used for identifying pluripotent stem cell residues are identified therefrom, and the marker gene is selected from the candidate genes for validation. The marker gene provided by the present invention can be used for detecting and identifying pluripotent stem cell residues in a derivative cell product of pluripotent stem cells for clinical treatment, so as to perform quality control on clinical cell products.
