Abstract:
Method and nucleic acids for the detection of microorganisms of the Yersinia pestis/Yersinia pseudotuberculosis species and/or differentiation between Yersinia pestis and Yersinia pseudotuberculosis in a sample. The invention aims to provide safe detection and differentiation methods for microorganisms of the Yersinia pestis/Yersinia pseudotuberculosis species. The method consists of several steps: (a) Bringing the sample into contact with a combination of at least two first nucleic acids (primers) which hybridize with an area of their microbial genome preserved in the species Yersinia pestis/Yersinia pseudotuberculosis or part thereof; (b) the amplification of the microbial genome or of a part of it to generate at least one amplification part; (c) bringing the amplification fragment of fragments obtained in step (b) into contact with at least one second nucleic acid (probe), which comprises a specific partial sequence of the amplificated genome or a part thereof for the speciesYersinia pestis/Yersinia pseudotuberculosis, wherein the second nucleic acid (probe) is specifically hybridized with at least one amplification fragment while forming at least one hybrid nucleic acid; (d) detecting the hybrid nucleic acid.
