Abstract:
Provided is a method for efficiently synthesizing, by a simple procedure, a double-stranded DNA library having different tagment sequences on the 5' terminal and 3' terminal sides. The method includes the following steps (i) to (iii): (i) a step for fragmenting a double-stranded DNA by using a transposome having (a) a first adapter containing a first tagment sequence and a first transposon-terminal sequence, (b) a second adapter containing a second transposon-terminal sequence having a base sequence complementary to at least a portion of the first transposon-terminal sequence, and (c) a transposase, and linking, as a transferred strand, either the first adapter or the second adapter to the 5' terminal or the 3' terminal of the double-stranded DNA; (ii) a step for linking an additional oligo containing a second tagment sequence to the second adapter; and (iii) a step for linking the non-transferred strand adapter, from among the first adapter and the second adapter, which is not linked to the double stranded DNA, to the end, among the 5' end and the 3' end of the double-stranded DNA, to which the transferred strand adapter was not linked in step (i).
