PROCESS FOR EXTRACTING SULFURIC ACID CONJUGATE AND GLUCURONIC ACID CONJUGATE OF STEROID

Abstract:
A process for extracting sulfuric acid conjugate and glucuronic acid conjugate of steroid, which comprises converting a sulfuric acid conjugate of steroid and a glucuronic acid conjugate of steroid in a sample solution (e.g., urine) to tetrapentylammonium ion pairs, first extracting the ion pair of the sulfuric acid conjugate with benzene or the like at a pH of 6 or more utilizing difference in extractability between the two ion pairs with organic solvents, adding sodium sulfate to the remaining sample solution in an amount of 20 wt % or more and, after dissolving the sodium sulfate, extracting the ion pair of the glucuronic acid conjugate with dichloromethane or the like at a pH of 7 or more. Also disclosed is another process for extracting the two ion pairs with dichloromethane or the like, which comprises adding 20 wt % or more sodium sulfate and extracting the ion pairs between the two conjugates and a tetraalkylammonium ion wherein the straight chain of the alkyl group contains 5 to 7 carbon atoms with dichloromethane or the like at a pH of 7 or more utilizing common extractability of the two ion pairs with organic solvents.
