Abstract:

A process for purifying a human BCDF by subjecting the reduced form of human BCDF, prepared by culturing a microorganism containing a human BCDF gene integrated therewith and solubilizing the culture product with guanidine hydrochloride, to oxidation and refolding, wherein the oxidation product is treated by gel filtration chromatography under the condition of the guanidine hydrochloride concentration of 4 to 7 M to thereby obtain a human BCDF having the intramolecular disulfide linkage and stereochemical structure of a natural human BCDF; a process for purifying a human BCDF by removing an organic solvent from the solution of a human BCDF containing said solvent, wherein the solution is passed through a gel filtration chromatographic column equilibrated with an organic solvent for elution according to the step-wise or linear gradient program to thereby obtain a natural human BCDF monomer; and a process for purifying a human BCDF which further comprises the steps of ion-exchange chromatography and reversed phase HPLC in the above processes. These processes permit the removal of impurities derived from the microorganism and human BCDF analogues, and the obtained natural human BCDF is highly pure so that it can be utilized as a medicine.
