PURPOSE: To remove factors caused by analyzing persons such as measuring errors, to reduce the amount of reagent and to make it possible to alleviate the loads of the analyzing persons by replacing the entire pretreatment with an autosampler having the pretreating function.
CONSTITUTION: Coumarin solution 4 of a standard sample whose concentration is known and n-dodecane 1 are measured by every specified amounts, mixed in a test tube 5, stirred and regulated. The measurement and the stirring are performed by sucking and discharging the sample with a syringe 8. The standard sample, n- dodecacane, alkali liquid 2 and alcohol liquid 3 after the preparation are mixed with reaction liquid 12. Air in a needle 10 is discharged into the alkali liquid at the lowermost layer. The alkali liquid is made to convect to the upper layer and stirred. The alkali liquid after the coumarin is extracted is distributed and diluted into the required quantity in a test tube 21. Then, the liquid is injected into a flow cell 13. Ultraviolet rays are emitted from a spectroscope 15 on the exciting side of the liquid, and isomerization is performed. Thereafter, the intensity of the coumarin fluorescence in the sample is detected 17 through a spectroscope 16 on the side of fluorescence. Operation 19 is performed, and a calibration curve is plotted and outputted 20. The same procedure is performed for the sample whose concentration is unknown. The concentration is computed based on the calibration curve obtained before.
