Abstract:

PROBLEM TO BE SOLVED: To obtain a new chimera protein containing a part rich in histidine residues at the N-terminal and capable of being easily cleave with a processing enzyme to efficiently give an objective protein as a substrate for the production of the physiologically active peptide (precursor).
SOLUTION: A new chimera protein of the formula: A-L-B {A is a protecting protein; B is an objective protein; L is a linker peptide; the C-terminal part has the sequence: X1-X2-(Pro, Lys or Arg)-Arg (X1, X2 are each an arbitrary amino acid); the N-terminal part has a His-enriched part such as the sequence: [(His)4-Pro-Gly]n [(n) is 1-6]}. As a substrate for producing the physiologically active peptide or its precursor, the chimera protein can easily be cleaved with a processing enzyme to efficiently give the objective protein. The chimeral protein is obtained by expressing a vector containing a gene bound to a DNA coding the protecting peptide, the linker peptide and the objective peptide, respectively, in a host cell.
