Abstract:

To provide a method for determining a specific oligonucleotide or polynucleotide sequence, capable of being easily and automatically carried out, not requiring the preliminary isolation of RNA, enabling the quantitative detection of nucleic acid and low in back ground signals.

This method for quantitatively detecting a specific nucleic acid sequence comprises hybridizing a specimen mixture containing an RNA or a single-stranded DNA with an oligonucleotlde probe or a polynucleotrde probe having a specifically bindable chemical group and a specifically detectable chemical group which are substantially complimentary with a polynucleotide sequence to be determined, adding an agent for cleaving a single-stranded polynucleotide sequence to the hybridization product, transferring the mixture to a proper reaction vessel, and subsequently detecting the immobilized nucleotide or non-immobilized nucleotide.
