Abstract:

PURPOSE: To easily determine the activity of interferon useful as an antiviral agent or an antitumor agent in improved sensitivity by reacting a specimen containing an interferon with a cell and determining the induced protein.
CONSTITUTION: A specimen containing interferon (e.g. interferon-3a or interferon-β) such as serum, blood plasma, buffer solution, medium or supernatant of cell culture medium is added to e.g. a buffer solution containing suspended cells such as leukocyte, monocyte, lymphocyte, fibroblast or epithelial cell or established cells such as THP-1 cell, U-937 cell, H9 cell, FL cell or WISH cell, incubating the mixture under a cell culture condition for 3-48hr and determining the induced protein composed of an enzyme such as 2'→5' oligoadenylic acid synthetase or phosphodiesterase to easily determine the biological activity of interferon in high sensitivity compared with conventional method.
