Abstract:

PURPOSE: To obtain a DNA having complimentarity to gene RNA of simian immunodeficiency virus by transforming E.coli with a plasmid, selecting a transformant and extracting the plasmid DNA from the transformant.

CONSTITUTION: λOngCSIVMND is digested with restriction enzyme XbaI and the obtained fragment is integrated into a plasmid having similar restriction enzyme site. An E.coli is transformed with the obtained plasmid and a transformant is selected by using the resistance to an antibiotic substance (e.g. ampicillin) as a marker. The selected transformant is subjected to alkali extraction to obtain a plasmid DNA integrated with the objective DNA having complimentarity to the RNA of SIVMND. The objective DNA having complimentarity to the RNA of SIVMND can be produced by digesting the plasmid with a restriction enzyme XbaI.
