Abstract:

To provide a dual controlled release matrix system based on a lipophilic vehicle suitable for controlled release of a low dose compound and a moisture-sensitive compound, and constituting a form in which an agent is hardly extracted from a dosage form.

A matrix is in the form of a suspension, enables easier formulation of a low dose compound and/or a moisture-sensitive compound, and includes two rate controlling barriers for controlled release of one or more pharmaceutically active materials. A first rate controlling barrier includes a relatively lipophilic oily vehicle, and can further include one or more excipients dissolved in the lipophilic vehicle and having itself a rate controlling property. A second rate controlling barrier is a hydrogel-forming polymeric material which is dispersed in the first rate controlling barrier. When the first rate controlling barrier breaks down, the pharmaceutically active agent is slowly released and a surrounding aqueous medium begins to ooze out into a polymer matrix.
