Abstract:

PROBLEM TO BE SOLVED: To obtain the subject new gene containing a DNA coding a polypeptide having endoglycoceramidase activator or functionally the same activity, useful for industrially producing an enzyme for hydrolyzing a sphingoglycolipid.
SOLUTION: This new endoglycoceramidase gene has a DNA coding a polypeptide having an amino acid sequence of formula I or formula II or a part of it and endoglycoceramidase activity or functionally the same activity and is useful for industrially producing a gene which hydrolyzes a glycoside bond between a sugar chain of sphingoglycoside and a ceramide to liberate the sugar chain of a complete form and the ceramide by enzyme gene manipulation technology. The gene is obtained by separating a genome DNA from Rhodococcus sp. M-777, a bacterium capable of producing endoglycoceramidase II, preparing a DNA library by using the genome DNA, screening the library by using a probe and recovering the DNA from a positive clone.
