CONSTITUTION: A hybrid promoter.operator (A) is obtained by containing a region having a base sequence expressed by formula I on the upstream (5') side of the -35th region with the upstream region of the base sequence as 0-15 bases, the -35th region having a base sequence expressed by formula II, the -10th region having a base sequence expressed by formula III at a distance of 15-25 bases from the -35th region to the -10th region and an operator part of a lactose operon at a distance of 0-15 bases from the downstream (3') side of the -10 region. The promoter part (B) in the resultant ingredient (A) is then constructed from a base seguence expressed by formula IV. The ingredient (A), as necessary, is subsequently introduced into a plasmid pUK02B10-pm4, etc., to afford a plasmid. Escherichia coli KY1436, etc., transformed with the resultant plasmid are then cultured to produce a human variant type prourokinase.
