A sample of a specimen containing protein originating from an erythrocyte is hemolyzed in a buffer soln. containing about 0.5-4% (w/v) of a surfactant such as carboxylate, sulfonate or sulfate to prepare a hemolytic sample. As a solvent of the buffer soln., water or a known buffer soln. may be used. At a time of immunoassay, the concn. of the surfactant is adjusted to about 0.01% (w/v) or less to prevent the suppression of antigen-antibody reaction. As a soln. at a time of immunoassay, a phosphate buffer soln., a tris-hydrochloric acid buffer soln. or a borate buffer soln. is used and, as immunoassay, an immunoagglutination method, a nephelometric method or enzyme immunoassay is used. A latex agglutination method is simple in operation and permits the measurement of many specimen samples for a short time.
